Overview

Microcephaly-deafness-intellectual disability syndrome (also known as Microcephaly-hearing loss-intellectual disability syndrome, or previously referred to as microcephaly-deafness-mental retardation syndrome) is an extremely rare genetic condition characterized by the combination of microcephaly (an abnormally small head circumference), sensorineural hearing loss (deafness), and intellectual disability. The syndrome primarily affects the central nervous system and auditory system, leading to significant developmental challenges from early life. Affected individuals typically present with congenital or early-onset microcephaly, reflecting reduced brain growth, which contributes to varying degrees of intellectual disability ranging from mild to severe. Sensorineural deafness, which results from dysfunction of the inner ear or auditory nerve pathways, is another hallmark feature and may be congenital or develop in early childhood. Additional features that have been reported in some cases include short stature, facial dysmorphism, and seizures, though the clinical spectrum may vary among affected individuals. This syndrome has been described in only a very small number of families in the medical literature, making it one of the rarest genetic conditions known. Due to its extreme rarity, there is no specific targeted treatment available. Management is supportive and multidisciplinary, focusing on early intervention programs, hearing aids or cochlear implants for hearing loss, speech therapy, special education services, and seizure management when applicable. Genetic counseling is recommended for affected families.
